Allele frequencies in a Gypsy Spanish population: Comparative analysis with other Spanish population databases.

  Gypsies represent about 0,6 % of the Spanish population. The study of some genetic markers and the estimation of the genetic distances show that Spanish Gypsies are closer to Indian Caucasoid populations than to the other non-Gypsy population.

 

The aim of this work is to establish a database for nine STR loci in a Gypsy population residing in the north of Spain (Vizcaya) and compare it with other Spanish available databases. Blood samples were taken from 151 unrelated Gypsy individuals. DNA extraction were performed with the commercial "blood high-genomic DNA purification kit" (Biotools-B&M Labs, S. A.). The co-amplification of D3S1358, vWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317 and D7S820 loci was carried out by PCR using a commercially available kit "AmlFl STR Profiler Plus PCR amplification kit" (Perkin-Elmer, Norwalk, CT, USA) and subsequently typed by capillary electrophoresis (ABI Prism 310 Genetic Analyzer).


  All the loci except D18S51, D3S1358 and vWA (c2 test) and D3S1358 and vWA (exact test) were in Hardy-Weinberg equilibrium. The results show large differences in allele frequencies among the Gypsy group and other Spanish population databases. In addition, we found in the Gypsy population some new allele not present in Spanish populations: the allele 33,1 in the D21S11 locus and the allele 23,2 in FGA locus.

  In conclusion, our results here presented suggest the usefulness of the construction of a particular database for these STR loci in this Gypsy population when these markers are u sed for forensic purposes.
